Title: IL-7 antagonizing the stimulating activity of TGF-β_1 to fibroblasts
Abstract: Objective The objective of this research is to make it clear whether Il-7 can antagonize transformation from lung-fibroblast to myofibroblast,fibroblast multiplication and generation of collagen under the stimulation of TGF-β1,and it can provide a new approach for treatment of lung fibrosis.Methods After the recovery of fibroblasts,under the stimulation of TGF-β1(concentration:10 ng/ml),the situation of cell multiplication at 24,48 and 72 hours(MTT evaluation),the collagen generation(hydroxyproline digestion examination)and α-SMA expression(western blotting examination)were observed.Then different concentrations of IL-7(50 ng/ml,100 ng/ml,150 ng/ml)were added to interfere at corresponding time,and they were reexamined after 24 hours.Results Different concentration of IL-7(50,100,150ng/ml)can inhibit the cell multiplication caused by TGF-β1(10 ng/ml)at different time(24 h,48 h,72 h).IL-7 with concentration of 50 ng/ml has no obvious influence on collagen generation of fibroblasts after stimulation of TGF-β1(10 ng/ml).The concentration of 100ng/ml IL-7 has influence on collagen generation by inhibition of its production and concentration of 150 ng/ml IL-7 has remarkable influence on inhibition of collagen production.Different concentrations(50 ng/ml,100 ng/ml,150 ng/ml)of IL-7 can reduce α-SAM expression of fibroblasts stimulated by TGF-β1,and its inhibitory effect depends on concentration.Conclusion This article confirmed that different concentrations(50 ng/ml,100 ng/ml,150 ng/ml)of IL-7 can reduce cell multiplication and α-SMA expression of fibroblasts under the stimulation of TGF-β1 in vitro.Different concentrations(100 ng/ml,150 ng/ml)of IL7 can reduce collagen formation of fibroblasts stimulated by TGF-β1 in vitro.
