Title: Dioscin stimulates proliferation and differentiation of MC3T3-E1 osteoblasts via upregulation of Lrp5 and β-catenin expression
Abstract: Aim To study the effects of dioscin on the proliferation,differentiation of MC3T3-E1 osteoblasts cultured in vitro and study the probable mechanism of dioscin on osteoporosis. Methods MC3T3-E1 osteoblasts cultured in vitro were treated by different concentrations of dioscin( 0. 3,0. 6,1. 2 μmol·L- 1) and lovastatin( 0. 04 μmol · L- 1) for different time respectively. The proliferation of osteoblasts was detected by MTT method. The differentiation of osteoblast was detected by the activity of ALP and Von Kossa staining. The expression of Lrp5,β-catenin,OPG and RANKL mRNA was examined by RT-PCR. RNA interference was used to detect whether Dioscin up-regulated the ratio of OPG / RANKL dependent on Lrp5 pathway. Results Dioscin stimulated the proliferation and increasing the ALP activity and the number of mineral nodes forming in cultured osteoblasts. At the same time dioscin also up-regulated the expression level of Lrp5,β-catenin mRNA and increasing the OPG/RANKL ratio. And the effect of dioscin increasing the OPG /RANKL ratio was blocked in the cells with Lrp5 knockout. Conclusions The suitable concentration of dioscin could promote proliferation, differentiation of MC3T3-E1 osteoblasts with a dose-dependent effect by upgrading the expression of Lrp5,β-catenin mRNA and increasing the OPG / RANKL ratio. And dioscin increasing the ratio of OPG / RANKL is dependent on the Lrp5 pathway.
