Abstract:Objective To study the effect and possible mechanism of dauricine on the differentiation and maturity of osteoblast(NHOst).Methods The normal human NHOsts were cultured in medium with 100ng/ml of daur...Objective To study the effect and possible mechanism of dauricine on the differentiation and maturity of osteoblast(NHOst).Methods The normal human NHOsts were cultured in medium with 100ng/ml of dauricine for seven days,the cell morphology was observed by phase contrast microscope,calcium deposition of the cells was detected by alkaline phosphatase(ALPase) and Von Kossa silver staining.The expression of core binding factor A1 mRNA(Cbfa1mRNA) was detected by RT-PCR.The expression of bone matrix protein osteocalcin was detected by Western-blot.Results After seven days cultivation,the cells grew in island shaped distribution and began to differentiate into mature cells in experimental group,mature cells formed bone nodules,but not formed bone nodules in control group.The calcium deposits were detected in experimental group,but not in control group by alkaline phosphatase and Von Kossa silver staining.The expression levels of core binding factor A1(Cbfa1) and osteocalcin(bone matrix protein) were higher in experimental group than in control group.Conclusion Dauricine can promote osteoblasts differentiation and maturity.Read More
