Title: Up-regulated expression of a prostate cancer-related gene,NSBP1, in prostate cancer cells
Abstract: To ascertain the expression level of nucleosomal binding protein gene (NSBP1) in prostate cancer cell lines. Five expressed sequence-tags chosen by mRNA differential display were used to screen the prostate cancer-related genes in the Non-redundant GeneBank of the National Center for Biotechnology Information (NCBI) by Blast program. The expression levels of NSBP1, the interested gene, was studied in the prostate cance cell lines using semiquantitative reverse transcription polymerase chain reaction(RT-PCR) and Northern Blot analysis. Four of five expressed sequence-tags expressed differentially in normal prostate and prostate cancer tissue failed to match in the Non-redundant GenBank, while the fifth one shared 97% homology with NSBP1. Compared with the normal prostate tissue, the expression levels of NSBP1 increased by 2.5-fold, 3.4-fold and 3.6-fold in LNCaP?DU145 and PC-3 cell line, respectively, which was in according with the results of Northern Blot,and the expression level of NSBP1 in 7 prostate cancer tissues was higer than that in normal ones. The paired-sample T test confirmed that the difference was significant statistically. The results suggest that the expression of NSBP1 was significantly higher in prostate cancer cell lines/tissues as compared with normal prostate tissue, which may be involved in carcinogenisis of the prostate cancer.
