Title: Effect of saponins from Panax notoginseng on pharmacokinetics of monoammium glycyrrhizinate in rats
Abstract: Objective:To study the pharmacokinetic features of monoammium glycyrrhizinate-Pa- Max notoginseng saponins(MG-PNS)compound as well as the effect of PNS on pharmacokinetics of MG. Methods:A high-performance liquid chromatographic method was developed to determine the plasma concentration of MG in the rats after bolus intravenous administration of MG solution or MG-PNS solution 20,50 or 100 mg·kg~(-1)(dose represented as MG),respectively.Pharmacokinetic parameters were calcu- lated with a 3P87 program.Results:The plasma MG concentration was biexponentially declined in both MG and MG-PNS groups,which obeyed a two-compartment phannacokinetic model.The MG pharmaco- kinetics featured nonlinearity.Compared to MG alone,the MG in the MG-PNS characterized significantly different pharmacokinetic profiles,e.g.,elevated AUC0_(0→∞)and reduced clearance rate at the dose of 100 mg·kg~(-1),and no significant fluctuations at the dose of 50 mg·kg~(-1). Conclusion:PNS affects the pharmaeokinetic profiles of MG in vivo in a dose-dependent setting.
