Abstract:Objectives To evaluate the practical value of PCR-SSP method for HLA class Ⅰ typing in cord blood samples.Methods T lymphocytes in 53 cord blood samples were isolated by T-immunomegnetic beads,and HLA...Objectives To evaluate the practical value of PCR-SSP method for HLA class Ⅰ typing in cord blood samples.Methods T lymphocytes in 53 cord blood samples were isolated by T-immunomegnetic beads,and HLA class Ⅰ were typed by serological method.The 38 cord blood samples were extracted for DNA,and were typed by PCR-SSP.Reliability,clinical practicability of both methods were compared according to the typing results.Results Serotyping and PCR-SSP typing for HLA-A have the same result,the discrepancy rate by serological method for HLA-B was 13.2%,including 1(1.4%) antigen being incorrectly interpreted and 9(11.8%) antigen of serologicalblanks turning out to be definable by PCR-SSP.The serological typing of cord blood should be tested within 24 hours,or else more than 50% of them can not be typed by serology.The 38 cord blood DNA were successfully extracted without time limitation.Conclusions PCR-SSP typing for HLA class Ⅰ is more accurate,reliable and blood-saving than serology,so it is suitable for typing of HLA class Ⅰ in cord blood bank.Read More
