Title: Pharmacokinetic study on grepafloxacin in rats
Abstract: Objective A sensitive, specific and rapid RP HPLC with fluorescence detection was developed and validated to determine grepafloxacin in rat plasma in order to study its pharmacokinetics in rats.Method The plasma samples were pre treated using simple liquid liquid extraction,with OPC 17203 being and internal standard.After oral administration of grepafloxacin (10 mg/kg) to rats, the concentrations in plasma were measured and main pharmacokinetic parameters were calculated by 3P87 program.Results and Conclusion The good linearity was achieved.ranging from 0 1 to 4 0 μmol/L,and the extraction recovery,accuracy and the precisions of intra day and inter day also were qualified.The main pharmacokinetic parameters were as follows: t 1/2 ( K a)=0 262 h, t 1/2 ( K e)=4 573 h, T max =1 211 h, C max =0 619 μmol/L,AUC= 4 860 h·μmol/L .
