Title: EGFR promoter methylation and mRNA expression in non-small cell lung cancer tissues
Abstract: OBJECTIVE: To investigate the relationship of epidermal growth factor receptor(EGFR) expression and histological type and lymph node metastasis as well as the correlation of EGFR promoter methylation and mRNA levels in non-small cell lung cancer(NSCLC) tissues.METHODS: The expression of EGFR was detected in 60 NSCLC samples using the immunohistochemical method.Methylation of EGFR and mRNA levels were detected by reverse transcription polymerase chain reaction(RT-PCR) and real-time fluorescence quantitative PCR.RESULTS: The positive rates of EGFR expression were 38.5%(10/26) in squamous carcinoma and 76.5%(26/34) in adenocarcinoma.Significant difference was present between them(χ2=8.87,P=0.002 9).The positive rates of EGFR expression were 73.7%(28/38) in patients with lymph node metastasis and 45.5%(10/22)in those without lymph node metastasis.The expression of EGFR in NSCLC was significantly correlated with lymph node metastasis(χ2=4.78,P=0.028).The EGFR mRNA expression of tumor tissues was higher than that in their adjacent mucosa in 11 cases of patients.In 12 cases of NSCLC,there was a negative correlation between EGFR promoter methylation and EGFR mRNA expression.CONCLUSIONS:The expression of EGFR has regulation with histological type of NSCLC and lymph node metastasis.The expression of EGFR mRNA is in negative regulation with the level of EGFR promoter methylation in some NSCLC patients.
