Title: Inhibition of Coxsackie virus B3 with aqueous extracts from Spatholobus suberectus in vitro
Abstract: Objective: To assess the effects and mechanisms of aqueous extracts from Spatholobus suberectus against Coxsackie virus B3(CVB3)in vitro.Methods:A series of concentrations of the aqueous extracts from Spatholobus suberectus Dunn.were incubated with CVB3-transfected Vero E6 cells.The survival rate of the Vero E6 cells by the aqueous extracts was detected by a MTT assay.The IC50 and therapeutic index(TI) of CVB3,CVB5,EV9,EV29 and PVⅠ were measured,respectively.Inhibition of CVB3-RNA replication by the aqueous extracts was assessed by a RT-PCR technique.Results: The aqueous extracts inhibited the cytopathic changes of the Vero B6 transfected by of the five species of Enterovirus in a dose-dependent manner,showing IC50 values of 60.8,47.1,17.1,65.5 and 29.1 μg·mL-1,respectively,and TI(CC50/IC50) of 4.1, 5.3,14.6,3.8 and 8.6,respectively.The aqueous extracts showed no inhibitions of CVB3 attachment onto Vero E6 but inhibited the RNA replication of the Vero B6 transfected by CVB3.Conclusion: Spatholobus suberectus inhibited the cytopathic changes by blocking the RNA replication of CVB3-transfected virus at early phase.
