Title: Influence of organic solvent on preparation of PLGA nanoparticles by emulsificationsolvent evaporation method
Abstract: Objective To prepare fluorescence marker coumarin-6 loaded nanoparticles by emulsification solvent evaporation method using the biodegradable material poly(lactic-co-glycolic acid)(PLGA) as carrier,and investigate the influence of organic solvent on nanoparticles′ formation.Methods Coumarin-6 loaded PLGA nanoparticles(coumarin-6-PLGA-NP) were prepared while organic phase was composed of dichloromethane(DCM) and ethyl acetate(EA) at different volume ratio.The entrapment efficiency and particle size of nanoparticles were determined by HPLC method and laser particle size analyzer.Results The nanoparticle size decreased with the increase of ethyl acetate,entrapment efficiency increased with the increase of dichloromethane.The particle size of nanoparticles and the entrapment efficiency was 128.7 nm and 35.9% respectively when combinations of DCM with EA were used at volume ratio of 70 ∶30.Conclusion Physical properties of organic solvents had significant effect on nanoparticles′ formation.PLGA nanoparticles prepared with combination of DCM and EA at ratio of 70 ∶30 exhibit narrow size distribution and higher entrapment efficiency.
