Title: Effect of Subacute Exposed to Dimethylformamide on Heart and Liver of Mice and Its Relationship with Lipid Peroxidation Level
Abstract: [Objective]To study the effect of subacute exposed to dimethylformamide (DMF) on heart and liver of ICR mice,explore its injury mechanism of lipid peroxidation.[Methods]40 mice were randomly divided into four groups,the mice in three dose group were poured into stomach through mouth with DMF at dose of 1.26,0.63 and 0.32 g/kg,and the control group was given normal saline,once a day for 50 days. After treatment period,the mice were sacrificed,the activity of alanine aminotransferase (ALT),aspartate aminotransferase (AST),lactate dehydrogenase (LDH),creatine kinase isoenzyme (CK-MB) and alpha-hydroxybutyrate dehydrogenase (α-HBDH) in serum were tested,and the contents of superoxide dismutase (SOD),malondialdehyde (MDA) and nitric oxide (NO) in tissues of liver and heart were detected. [Results]After 50 days of treatment,the levels of ALT and AST in serum and SOD and MDA in liver tissue didn't changed significantly,and the contents of NO in the low dose group was lower than that in the control group significantly (P0.05). There was no significant different in levels of LDH、α-HBDH in serum among these groups,but the levels of CK-MB in the high dose group and the low dose group were higher than those in the control group and the medium dose group significantly (P0.01,P0.05). There was no significant different in contents of SOD and NO in heart tissue among these groups,but the contents of MDA in treatment groups increased,and there were significant different in that between the medium dose group,the low dose group and the control group (P0.05 ,P0.01).[Conclusion]At the dose of this experiment,DMF had no significant effect on mice's livers,but it caused damage on mice's hearts. DMF can increase the level of myocardial lipid peroxidation,there is relationship between myocardial injury and increase of lipid peroxidation probably.
