Title: Effect of combined use of rAe-p53 and DDP in different time sequence on tumoricidal activity against human lung adenocarcinoma cells in vitro
Abstract: Objective:To investigate the best time sequence combined treatment with rAd-p53 (Gendicine) and cisplatin(DDP) to the lung adenocarcinoma cells in vitro. Methods:Inhibition ratio to human lung adenocarcinoma cells (A549) of rAd-p53 combined with DDP in different time sequence was assessed by the method of MTT and change of cell cycle was detected by flow cytometry (FCM). Results:rAd-p53 combined with DDP showed a notable addition or synergistic effect (Q0.85 or Q 1.15). There were statistical significances between combined drug groups and single drug groups (P0.01). The inhibition ratio of the group combined in different time sequence were all higher than that of the group single drug in same time (P0.01). There was the highest Q value at the time DDP was added after the time rAd-p53 was applied 48h (Q=1.38). This time A549 cells were more arrested in G0/G1 and cells in S phase significantly decreased. Conclusion:Combined treatment of rAd-p53 and DDP showed a notable synergistic effect. The best time sequence of combined treatment is rAd-p53 administered before DDP 48h and the mechanism is associated with cell cycle.
