Title: Effect of Different Polar Extracts of Uighur Ocimum basilicum L. on COX-2 Enzyme Activity in LPS-induced RAW264.7 Cells
Abstract: Objective To study the effect of Uighur Ocimum basilicum L.on COX-2 enzyme activity in LPS-induced RAW264.7 cells,and find its anti-inflammatory mechanism and effective parts.MethodsMTT method was used to the eligible concentration of different polar extracts of Uighur Ocimum basilicum L..After RAW264.7 were interfered with 2 mg·L-1 LPS for 8 h,and then different polar extracts of Uighur Ocimum basilicum L.for 30 min,the concentration of PGE2 in supernatant was analyzed by ELISA.After RAW264.7 cells were pretreated with different polar extracts of Uighur Ocimum basilicum L.for 30 min,and then 2 mg·L-1 LPS for 24 h,the concentration of PGE2 in supernatant was analyzed by ELISA.ResultsThe concentration of PGE2 was inhibited by different polar extract of Uighur Ocimum basilicum L..The inhibition ratio by acetidin part and n-butanol part were higher.ConclusionThe anti-inflammatory activity of Uighur Ocimum basilicum L.was related to inhititing the activity of COX-2 enzyme,and the effective parts are acetidin part and n-butanol part.
