Title: Downregulation of HDPR1 is associated with malignant phenotype of human lung cancer
Abstract: Objective To study the expression of HDPR1(human homologue of Dapper)in nonsmall cell lung cancer(NSCLC)and analyze if the expression is correlated with β-catenin and p120-catenin(p120ctn)expressions and malignant degree.Methods Immunohistochemistry S-P method,RT-PCR and western blot were used to detect the expressions of HDPR1,p120ctn and β-catenin in lung cancer tissues.Results Immunohistochemical analysis showed that HDPR1 expression level was significantly higher in normal lung tissue than in NSCLC tissues(82/120,68.3%),and reduced HDPR1 expression in NSCLC tissues was related to the clinicopathological factors and poor prognosis of the patients with NSCLC.In addition,reduced HDPR1 expression was positively correlated with the decreased expression of p120ctn,and negatively correlated to increased expression of β-catenin in lung cancer tissues.Moreover,RT-PCR and western blotting analysis showed that the expression of HDPR1 mRNA and protein was lower in tumor tissues as compared to corresponding nontumorous tissues.Conclusion The downregulation of HDPR1 expression is associated with the abnormal expression of p120ctn and β-catenin,and correlated with the malignancy degrees of human lung cancer.
