Title: Relationship between nm23-H1, proliferating cell nuclear antigen expression and DNA content of human hepatocellular carcinoma
Abstract: Objective To evaluate the relationship between expressions of nm23 H1, proliferating cell nuclear antigen (PCNA) in hepatocellular carcinoma and DNA content of cancer cells. Methods The expression of nm23 H1 and PCNA were detected in 56 cases of HCC by using immunohistochemistry technique and DNA content of cancer cells were analyzed by DNA imaging analyses system as well in order to find their relationship with biopathologic characters of HCC. Results The expression levels of nm23 H1 in HCC without encapsulation (29.6%) were significantly reduced when compared with that with encapsulation (64.3%) or with incomplete capsulation (66.7%, P 0.05). The increased DNA content of HCC was correlated with non encapsulation, compact type and high grade of the tumor ( P 0.05). The labeling indexes of PCNA in the group with negative nm23 H1 expression of HCC were significantly higher than those in the group with positive nm23 H1 expression of HCC ( P 0.05). The DI in high proliferating group of HCC(2.30±0.90) were significantly higher than in low proliferating group (1.86±0.70, P 0.05). Conclusion The expression of nm23 H1 in HCC was correlated with formation of tumor capsulation and cancer cell proliferating activity. The analysis of DNA content of tumor cells in combination with the immuohistochemistry staining of PCNA could give accurate index for the malignant biopathologic characters of HCC.
