Title: Effect of zinc on apoptosis and P53 et al gene expression in HL-60 cells
Abstract: Objective To study the effect of trace element zinc on apoptosis in HL-60 cells and its mechanism.Methods The HL-60 cells were cultured by usual rules for(24?h) and then divided into group of control,N,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine(TPEN).TPEN+ZnSO_4.The apoptosis of cells was observed by means of morphological changes,flow cytometry analysis and agarose gel electrophoresis of DNA.The genes expression associated with apoptosis were detected in both mRNA and protein levels by mRNA slot-blot and western-blot.Results Intacellular Zn~(2+) decrease could induce apoptosis in HL-60 cells and down-regulate expression of genes bcl-2,c-myc in both mRNA and protein levels,but additiion of ZnSO_4(final concentration of(20?μmol/L))improved the conditions.Conclusion Down-regulated expression of genes bcl-2,cmyc are associated with the process of apoptosis induced by intracellular Zn~(2+) decrease.
