Abstract:Objective To investigate the role of Ca 2+ preconditioning in the mechanism of the myocardial endogenous protective effects.Methods Fifty six Sprague Dawley male rats were divided into 7 groups random...Objective To investigate the role of Ca 2+ preconditioning in the mechanism of the myocardial endogenous protective effects.Methods Fifty six Sprague Dawley male rats were divided into 7 groups randomly:IC group,ischemic control;CPC group,hearts being perfused for 3 cycles of Ca 2+ free and Ca 2+ containing KH buffer;IPC group,ischemia followed by reperfusion; CPC+Verapamil group;IPC+Verapamil group;Bay K8644 group;Verapamil group.Heart function and coronary flow were examined before ischemia and after reperfusion.The release of LDH and the content of ATP were investigated and ultrastructural alterations in the myocardium were examined.Results Significant heart functional recovery rate was observed in IPC and CPC groups as compared with that in IC group(P0.05).Administration of verapamil completely abolished the IPC and CPC induced cardiac protection.Administration of Bay K8644 could mimic cardiac protection against ischemia reperfusion injury.Conclusion IPC and CPC can induce myocardial endogenous protective effects against ischemia reperfusion injury.Administration of verapamil reduces Ca 2+ influx and can mimic cardioprotection against ischemia reperfusion injury.Read More
