Title: Effects of Shenqi Dihuang Decoction on Nephrin,Podocin,Nephrin mRNA and Podocin mRNA in Renal Tissue of IgA Nephropathy Rats
Abstract: Objective To observe the effect and possible mechanism of Shenqi Dihuang Decoction[decoction of Dangshen(Radix Codonopsis),Huangqi(Radix Astragali)and Dihuang(Radix Rehmanniae Recens)]on IgA nephropathy.Methods Sixty SD rats were randomized into normal group,model group,tripterygium group,low-dose Chinese medication group and high-dose Chinese medication group,with 12in each.The IgA nephropathy rat models were established with combined method [bovine serum albumin(BSA)+lipopolysaccharide(LPS)+ CCl4].The low-dose Chinese medication group and high-dose Chinese medication group was given 1.4g/100g and 2.8g/100g Shenqi Dihuang Decoction intragastrically.The tripterygium group was given 0.625mg/100g tripterygium wilfordii tablets intragastrically.The model group and normal group were given normal saline intragastrically once daily.The rats were killed at the end of 12weeks.The serum creatinine(Scr),urinary protein excretions(UPEs)and serum albumin levels were detected.The expressions of Nephrin and Podocin in renal tissues were detected with immunohistochemistry and Western-blot method.The expressions of Nephrin mRNA and Podocin mRNA in renal tissues were detected with reverse transcription-polymerase chain reaction(RT-PCR).Results Comparing with the normal group,the UPEs in other groups were significantly increased(P0.05).Comparing with the model group,the UPEs in three administration groups were significantly decreased(P0.05).Comparing with the normal group,the expressions of Nephrin,Podocin,Nephrin mRNA and Podocin mRNA in renal tissues in other groups were significantly decreased(P0.05).Comparing with the model group,the expressions of Nephrin,Podocin,Nephrin mRNA and Podocin mRNA in renal tissues in three administration groups were significantly increased(P0.05).Conclusion Shenqi Dihuang decoction can effectively decrease the urine protein of IgA nephrophaty rats,which may be relevant to up-regulating the expressions of Nephrin and Podocin in the kidney tissue.
