Title: Genetic diversity analysis of cultivated and wild populations and family groups of Siniperca chuatsi
Abstract: 5 microsatellite DNA markers were selected to analyze the genetic structure of three Siniperca chuatsi population progenies:Hubei,Guangdong cultured population and Yangtze River wild population and family groups.Optimum breeding pairs could be selected by means of microsatellite to evaluate the heritability of reproductive populations.5 microsatellite loci were highly informative,and the average polymorphic information content(PIC) value0.5.The genetic diversity among three populations was:Hubei(BF)Yangtze River(WBF)Guangdong(DF).The values of expected heterozygosity(He) in different family groups were between 0.4-0.7 and genetic differentiations between family groups were significant.Analysis of molecular variance(AMOVA) revealed that most(83.33%) of the total variation was within populations.Based on these observations above,the Hubei populations showed a relatively higher genetic diversity and selection of excellent populations from these groups was possible.The microsatellite molecular marker technology can be used for parental identification in later S.chuatsi breeding.
