Title: Function, expression and significance of TR6 in breast cancer
Abstract: Objective To study the function and expression of TR6 in breast cancer and its clinicopathological significance. Methods The protective effect of proliferation in breast cancer cell line MDA435 was studied by Elisa-like and 3H-TdR incorporation methods. Immunohistochemistry was performed to detect TR6 expression in 29 cases of breast carcer, 20 cases of the cancer metastases in lymphnode and 4 cases of breast benign lesion. Western blot was used to detect TR6 expression in 5 cases of breast cancer with available fresh tissue and breast cancer cell lines ML-20 and MDA435. Then the relationship between expression and clinicopathological features was analysed. Results Addition of Fas-L and TR6 had enhanced effect on proliferation of breast cancer cell line, while using TR6 antibody to neutralize TR6, the effect of proliferation was decreased. Immunohistochemistry showed that benign breast lesions did not express TR6 (0/4), and the expression of TR6 was detected in 18 in 29 cases (62%) of primary breast cancer, in which primary breast cancer without lymph node metastasis expressed TR6 in 4/9 (44%) cases, and that with lymph nodes metastasis in 14/20 (70%); and TR6 expression was detected in 16/20 cases (80%) of metastatic breast cancer. Conclusion TR6 has significantly enhanced effect on proliferation of MDA435 breast cancer cells line. One of the main mechanism may be that TR6 blocks the Fas-L-induced apoptosis. In comparison with benign lesion, there is more expression in primary and especially in metastasis of breast cancer tissue. TR6 may be a new target for early diagnosis and therapy of breast cancer.
