Title: Expression of claudin-1 in cervical lesions and its diagnostic value
Abstract: Objective To detect whether the expression of claudin_1 is associated with the progression of uterine cervical neoplasia and to investigate the diagnostic value of claudin_1 in cervical lesions. Methods We chose 105 formalin_fixed paraffin embedded cervical tissues that included 16 cases of normal cervical epithelium,21 mature squamous mataplasia or reparative epithelium, 21 low_grade squamous intraepithelial lesion (LSIL),27 high_grade squamous intraepithelial lesion (HSIL) and 20 invasive squamous cell carcinoma (ISCC). 77 cases of cervical tissues were constructed into tissue microarrays and the others were cut with the full tissue sections. Then, the expression of claudin_1 was determined by immunohistochemistry. Results The subcellular location of staining was membrance and/or cytoplasm.In the normal squamous epithelium, only the cells of parabasal layer were weakly stained.However, claudin_1 was increasingly expressed in squamous mataplasia or reparative epithelium, LSIL, HSIL and ISCC. The discordance between squamous mataplasia or reparative epithelium and neoplasia lesions had no statistical significance (P 0.05). Conclusions The expression of claudin_1 may be helpful for distinguishing hyperplasia from normal cervical epithelium, but it can not distinguish squamous mataplasia or reparative epithelium from neoplasia lesions. Therefore, our results support that claudin_1 can serve as a marker for the detection of cervical epithelium hyperplasia.However, further studies should be done to investigate whether claudin_1 can serve as a marker for the detection of cervical neoplasia lesions or not.
