Title: The immunological effect of Ag85A and Ag85B DNA vaccine on rat bladder cancer
Abstract: Objective To explore the immunological effect of the DNA vaccine encoding mycobacterium tuberculosis Ag85A with Ag85B in mice with bladder cancer. Methods The mice with bladder cancer were constructed by injecting mice bladder cancer cell strain BTT739 subcutaneously,which were then divided randomly into six groups:NS,BCG,pcDNA3.1,pcDNA-Ag85A,pcDNA-Ag85B,and pcDNA-Ag85A+ pcDNA-Ag85B groups. The mice were injected subcutaneously on the 7th,14th,and 21st day respectively. After 7 days,the number of CD4+ and CD8+ T cell subsets was detected,the IFN-γ in mice serum assayed. Finally,the tumor was weighed and studied histopathologicaly. Results The number of CD4+ and CD8+ T cell subsets,the ratio of CD4+/CD8+ and the IFN-γ level of the mice serum in Ag85A and Ag85B DNA vaccine groups,were significantly higher than those in the DNA vaccine alone group. The weight of tumor in Ag85A and Ag85B DNA vaccine group was less than in the DNA vaccine alone group (P0.05),and more immunocytes in tumor tissue and peri-tumor were observed. The immunological effect of Ag85A and Ag85B DNA vaccine group,pcDNA-Ag85A group,pcDNA-Ag85B DNA group was lower than that of BCG group. Conclusion Ag85A and Ag85B DNA vaccine have stronger immunization effect on the bladder cancer than DNA vaccine alone,which have synergetic effect after joint application,but their immunization effect is still lower than that of BCG.
