Abstract:ObjectiveTo study the determination method of puerarin and isoflavones in Pueraria lobata(Willd)Ohwi and Pueraria thomsonii Benth.MethodsThe separation was performed on RP-HPLC with a mobile phase of ...ObjectiveTo study the determination method of puerarin and isoflavones in Pueraria lobata(Willd)Ohwi and Pueraria thomsonii Benth.MethodsThe separation was performed on RP-HPLC with a mobile phase of CH3OH-H2O(32∶68V/V) solution and the detection wavelength was 250 nm for the determination of Pueraria.UV-spectrophotometry for the determination of isoflavones.ResultsPuerarin and other peaks were separated well and the peak shape was presented well.There was a good linearity within the range of 0.204 to 2.04 μg of Puerarin(r =0.999 9).The average recovery was 98.7 %(n=5).The RSD was 1.3%.The isoflavones had a good linearity within the range of 4.08~16.32 μg/ml(r=0.999 8).Conclusion This method is sensitive,accurate and reproducible.It can be used as the quality control method of Pueraria lobata and drug.Read More
