Title: The Hydrolysis Condition of Sand Jellyfish ACE Inhibitory Peptide Preparation
Abstract: In this paper,the preparation of ACE inhibitory peptide made from fresh sand jellyfish using the alkaline protease enzyme is studied. Using the degree of hydrolysis(DH) and ACE inhibition rate as indicators,the single factor experiments and orthogonal experiment are done to get the best hydrolysis conditions: reaction temperature 50 ℃,enzyme dosage of 5%,enzymatic hydrolysis starting pH 9.5. The improved high performance liquid chromatography method is used for the determination of ACE inhibition rate,and found that the maximum rate of ACE inhibition is get after hydrolyzed 7 h in the above condition. Through desalted and lyophilized,the inhibition of 0.5 mg/mL sand jellyfish ACE peptide is 54.53%. Finally,the average molecular weight of the obtained sand jellyfish ACE peptide is measured by exclusion chromatography with about500 Da.
