Abstract:Objective Edaravone,as a free radical scavenger,is used clinically to treat acute brain infarction,and its antioxidation may protect the lungs against injury.This study evaluated the protective effect...Objective Edaravone,as a free radical scavenger,is used clinically to treat acute brain infarction,and its antioxidation may protect the lungs against injury.This study evaluated the protective effect of edaravone against lung injury induced by acute paraquat intoxication in rats.Methods Seventy-two Sprague-Dawley rats were randomly divided into a blank control group(n=8),given normal saline,a paraquat group(n=32),injected intraperitoneally with paraquat at 18 mg/kg without further treatment,and an edaravone group(n=32),treated by daily intraperitoneal injection of edaravone at 6 mg/kg for 7 days immediately after paraquat intoxication.At 1,3,5 and 7 days after the treatment,the three groups of rats were detected for the contents of malondialdehyde(MDA),interleukin 6(IL-6) and tumor necrosis factor alpha(TNF-α) as well as the activity of superoxide dismutase(SOD) in the lung homogenate and bronchoalveolar lavage fluid(BALF) and that of glutathione peroxidase(GSH-PX) in the lung homogenate.The gross appearance of pulmonary injury was observed,and so were the pathological changes of the lung tissue under the light microscope.Results Compared with the blank control group,the contents of MDA,IL-6 and TNF-αwere markedly increased(P0.05 or P0.01),while the activities of SOD and GSH-PX significantly decreased(P0.01) in the lung homogenate and BALF of the paraquat group.The edaravone group showed significantly decreased contents of MDA,IL-6 and TNF-α(P0.05 or P0.01) and increased activities of SOD and GSH-PX in the lung homogenate and BALF(P0.05 or P0.01) as compared with the paraquat group.Lung injuries were remarkably reduced and no fibrosis observed under the light and electron microscopes after edaravone treatment.Conclusion Edaravone,as a novel free radical scavenger capable of lessening paraquat-induced lung injury,inhibiting lipid peroxidation,reduce the level of some inflammation media in the early stage of the injury,and abating diffuse pulmonary damage,promises to be a new medicine for acute paraquat intoxication.Read More
