Title: CFSE and PI two fluorochrome stain assay for detecting cytotoxicity
Abstract: Objective To investigate the assay of detecting cytotoxicity by 5-(6)-carboxy-fluorescein succinimidyl ester (CFSE) and propidium iodide (PI) two fluorochrome staining. Methods CFSE was used to label target cells and PI was used to stain killed cells. Splenocytes of immuned C57BL/6 and dexamethasone treated C57BL/6 were harvested for effector cells, and splenocytes of Balb/c were labeled with CFSE for target cells. Effector cells and target cells were mixed and cultured for 6 hours, and then were stained by PI. Fluorescent strength of cultured cells was measured by fluorescence activated cell sorting (FACS). Results CFSE and PI labeled splenocytes could clearly detected by FACS. CFSE and PI double positive cells were the killed target cells. There was a higher cytotoxic activity of immuned group than dexamethasone treated group(P 0.05). Conclusion CFSE and PI fluorochrome stain assay is simple and convenient for cytotoxicity detecting.
