Abstract:Objective To investigate the effects and its mechanisms of curcumin derivatives(Curc-OEG) on experimental rat hepatic fibrosis.MethodsMale SD rats were divided into four groups :normal group,model gro...Objective To investigate the effects and its mechanisms of curcumin derivatives(Curc-OEG) on experimental rat hepatic fibrosis.MethodsMale SD rats were divided into four groups :normal group,model group,curcumin derivatives group and curcumin group.Rat model of hepatic fibrosis was induced by CCl4,and then the model rats given with intravenous injection of 100 mg·kg-1 Curc-OEG and 400 mg·kg-1 curcumin.After treatment for 10 weeks,liver function,serum markers of hepatic fibrosis,liver Hpy content level were detected.The HE and Sirius red staining of hepatic fibrosis were observed and the progression of hepatic fibrosis was to grade,immunohistochemical for TGF-β1 and α-SMA in rat liver tissue were distributed.The liver tissue of TGF-β1 and Collagen I mRNA levels were detected by RT-PCR.The protein expression of TGF-β1 and α-SMA were detected by Western blot.ResultsCompared with model group,Curc-OEG could improve liver function and reduce serum levels of liver fibrosis,decrease levels of liver Hpy content,inhibitting the expression of cytokine TGF-β1 and α-SMA in liver tissue.ConclusionThe Curc-OEG has a significant effect of anti-liver fibrosis.Read More
