Title: Inhibitory Effect of Compound Liuyueqing on Synthesis of Collagen Type I in Hepatic Tissues in Rats with Hepatic Fibrosis
Abstract: Objective: To study the inhibitory effect of Compound Liuyueqing(CLYQ) on the synthesis of Collagen type I(Col I) in hepatic tissues in rats with hepatic fibrosis.Method: Hepatic fibrosis model of SD rats was induced by subcutaneous injection(sc) of CCl4.Hepatic fibrosis SD rats were divided into 5 groups randomly: model control group,colchicina-group,high dose,middle dose and low dose of CLYQ groups,and normal SD rats were the blank contol group.Model control group and blank contol group were treated with normal saline,and the other groups were treated with corresponding drugs for four consecutive weeks by intragastric administration(ig).The relative quantification of Col I mRNA expression in hepatic tissues was detected by real time fluorescence quantitative PCR,and the quantification of Col I protein expression was detected by immunohistochemistry.Result: Compared with the model group,the various dosage groups of CLYQ could down regulate significantly Col I mRNA expression(P 0.01) and showed a dose-dependent relationship according to the results of real time fluorescence quantitative PCR.The results of immunohistochemistry showed that after treamented with CLYQ the area and the intensity of Col I protein expression in hepatic tissues were reduced and weakened significantly(P 0.05,or P 0.01) compared with that of the model group.Col I protein expression was reduced obviously in a dose = dependent manner.Conclusion: CLYQ can down-regulate the expression of Col I mRNA and protein synthesis in hepatictissues of hepatic fibrosis rats.The results suggest that CLYQ can reduce the collagen,thus suppressing the deposition of ECM.Hence CLYQ has the marked effect of anti-hepatic fibrosis.
