Abstract: The effect of ascorbic acid on hemolysis induced by hemin was investigated. The degree of hemolysis was determined spectrophotometrically by the concentration of hemoglobin in the supernatant at 540 nm.Thiobarbiyuric acid reactive substrates(TBARS) in erythrocytes were measured by the method established by Stocks et al.Methemoglobin in erythrocytes was determined by the method reported by Sezebeni et al.The current results demonstrated that ascorbic acid enhanced hemin-induced hemolysis.Although hemin itself did not induce formation of TBARS and methemoglobin in erythrocytes,combination of hemin and ascorbic acid promoted formation of TBARS and methemoglobin in erythrocytes.Catalase abolished the addition hemolysis as well as TBARS and methemoglobin mediated by hemin and ascorbic acid.Several HO~ scavengers significantly inhibited hemolysis induced by hemin and ascorbic acid.It was concluded that H_2O_2 was involved in the oxidative stress mediated by hemin and ascorbic acid.HO~ might be the cause of ascorbic acid-enhanced hemolysis induced by hemin.Ascorbic acid could function as a pro-oxidant,rather than an antioxidant,in the presence of hemin.
