Title: Antioxidant effect of scutellarin on PC12 cell injury induced by hydrogen peroxide in culture
Abstract: PurposeTo investigate the antioxidant effect of scutellarin on PC12 cell injury induced by hydrogen peroxide in culture.MethodsThe PC12 cells injury was induced by H_2O_2 in culture.Morphological changes of PC12 cells were visualized under light microscope.Lactate dehydrogenase(LDH),malondialdelyde(MDA) and superoxide dismutase(SOD) in the culture medium and MDA,SOD in the cell were measured by spectroscopy respectively.ResultsAfter being exposed to 100 μmol/L H_2O_2 for 4 hours,typical morphological characteristics of injury were observed.The level of LDH and MDA was increased significantly while SOD level was decreased significantly in H_2O_2 induced group compared with that in the sham group.Scutellarin can decrease LDH,MDA content and increase SOD activity significantly.ConclusionScutellarin has protective effect on PC12 cell injury induced by hydrogen peroxide in culture.The protective effect may be related to protecting cell membrane,to increasing the activity of clearance enzyme of free radical and to inhibiting lipid peroxidation.
