Title: The Effect of Genistein on the Proliferation and Cell Cycle of Human Breast Carcinoma Cell MDA-MB-435S
Abstract: Objective: To investigate the effect of genistein on the survival rate, cell cycle, and apoptosis of breast carcinoma cell MDA-MB-435S cultured in vitro. Methods: The effects of genistein on the growth of breast carcinoma cells were detected by MTT assay; the relation between the alteration of cell cycle of MDA-MB-435S and different dosage and duration of genistein was observed by flow cytometry, and the action of genistein on apoptosis of MDA-MB-435S cells was observed by fluorescent microscope with acridium orange/ethidium bromide staning. Results: The more inhibitory action of genistein was on the proliferation of MDA-MB-435S cell line, the more dosage and duration of genistein was. Furthermore, genistein could block the cell cycle from G2 to M stage, and the blockage was closed to the dosage and duration of genistein. The cell apoptosis induced by genistein also depended on the dosage. Conclusion: Genstein may inhibite the proliferation of human breast carcinomar cell MDA-MB-435S line in vitro, and genstein may play a role in apoptosis and blocking cell cycle of MDA-MB-435S cell line cultured in vitro.
