Title: Determination of gallic acid content in Chinese olive flesh by RP-HPLC
Abstract: A reverse-phase high-performance liquid chromatography(RP-HPLC) method was used for measuring gallic acid content in flesh of 15 Chinese olives varieties.Samples were injected onto reversed-phase Nucleosil C18 column(250 mm×4 mm i.d.).Mobile phase A and B were 20% methanol and 30% acetonitrile respectively.Both of them were adjusted to pH3.2 with 50% phosphoric acid.Acetonitrile was set as mobile C.Triple gradient elution proceeded at the flow rate 0.8~1 mL/min.Operation conditions were as follows: column temperature 35 ℃,detection at 280 nm of wavelength.The results showed that linear calibration of gallic acid was within 0.390 6~25.000 0 μg/mL with correlation(r)of 0.999 6.Regression equation was acquired as y=62.662x-16.805.Relative standard deviation(RSD) of retained time and peak area of standard sample were respectively 1.761 4% and 2.773 6%.Average recovery of standard sample was 99.09%(n=5) with RSD of 3.298%.Gallic acid was detected in all olive samples with the content within 57.958 4~230.737 3 μg/g fresh mass(FM).According to the content of gallic acid,the richest variety was Tunatianzhong while the least was Xiangzhong with RSD of 0.060%~1.938%.
