Title: Anti-angiogenic mechanism of all-trans-retinoic acid in vitro
Abstract: Objective: To investigate the effects of all-trans-retinoic acid (ATRA) on the endothelial cell and elucidate the anti-angiogenic mechanism of ATRA. Methods: MTT assay, cell migration assay and in vitro angiogenesis assay were applied to determine the effects of ATRA on the proliferation, migration and angiogenic ability of endothelial cells respectively, also we detected the apoptosis of endothelial cells induced by ATRA through flow cytometry analysis. Results: The proliferation, migration and angiogenic ability of endothelial cells were suppressed by ATRA, the number of the apoptotic cells increased significantly in the groups treated with ATRA compared with the control ones. Conclusions: ATRA may inhibit the angiogenesis through its effects on endothelial cells directly, and the suppression of proliferation, migration, angiogenic ability of endothelial cells and the induction of apoptosis are the possible action mechanisms.
