Title: Protective effects of Curcumin on the lung ischemia-reperfusion injury after rat unilateral lung orthotopic transplantation
Abstract: Objective To investigate the changes in lung function of ischemia-reperfusion injury (IRI)after rat unilateral lung orthotopic transplantation and the protective effects of CUR.Methods Rats were subjected to unilateral left lung orthotopic transplantation.A total of 120 male inbred Sprague-Dawley rats were randomly divided two groups,and each group was subdivided into sham,vehicle,CUR group (n=12/per subgroup).The animals were sacrificed at ischemia period of 4 and different reperfusion peri- eds of 2 h and 24 h,respectively.PO_2/FIO_2 ratio,lung injury score,wet/dry ratio(W/D),and evan's blue dye(EBD)in transplanted lungs were observed.Results After lung reperfusion,the histological ap- pearances were mainly presented with pulmonary edema,inflammatory cell infiltration and hemorrhage.As compared with sham group,after 2 h of reperfusion,the PO_2/FIO_2 ratio was reduced significantly from 358. 3 to 153.7;the W/D was increased significantly from 3.7 to 5.6;and the EBD content was increased sig- nificantly from 381.0μg/g dry weight to 1172.3μg/g dry weight in the transplanted lungs.After 24 h of reperfusion,the PO_2/FIO_2 ratio was reduced significantly from 394.2 to 102.3;the W/D was increased significantly from 3.8 to 6.6;and the EBD content was increased significantly from 387.5μg/g dry weight to 927.5μg/g dry weight in the transplanted lungs as compared with those in the sham group.Pre- treatments of the donor and recipient rats with CUR could significantly reduce the lung injury score,the alveolar-capillary leakage,W/D,and improve gas exchanges in animals submitted to 4 h of ischemia com- bined with 2 h or 24 h of repeffusion.Conclusion The lung function changes of IRI after rat unilateral lung transplantation mainly were permeability lung edema caused by alveolar capillary membrane injury and the reduction of lung oxygenation function.Cureumin attenuated IRI after experimental lung transplan- tation.
