Title: Identification of phages expressed β-amyloid-targeting peptide using phage display random peptide library
Abstract: Objective To screen the targeting peptides of β-amyloid(Aβ) for attenuation of the neural toxicity of Aβ. Methods Aβ1-10 was synthesized and labeled by biotin. A randomized 12-mer peptide library presented on M13 phages was used to screen the peptides(phages) specifically bond to the bio-Aβ1-10. The phages specifically binding to the bio-Aβ1-10 was examined by BIAcore3000 instrument. The affinity and kinetics of the interaction between the screened phages and the target molecule were detected by binding specificity test of the screened phages and competitive inhibition test of the small peptides using bio-sensing techniques. Results The phages with binding affinity for bio-Aβ1-10 were acquired after 3-round screen. Bio-sensing technique showed high binding specificity between the selected phages and bio-Aβ1-10. Conclusion The phages with high affinity and binding specificity with Aβ1-10 can be obtained by using bio-sensing techniques.
