Title: Apoptosis and cell proliferation of colon carcinoma Lovo cells after transient transfection siRNA targeting survivin gene
Abstract: Objective To introduce survivin siRNA to colon carcinoma Lovo cells and study the influence on apoptosis and cell proliferation. Methods Two pairs of siRNAs specific targeting survivin gene were designed, synthesized by vitro transcription, and transfected into Lovo cells by lipofectamine 2000. The cell apoptosis and cell proliferation were detected. Results Survivin mRNA expression level was detected with RT-PCR. Two survivin-targeted siRNA groups' survivin mRNA expression levels were both lower than normal Lovo's. The cell apoptosis of two survivin-targeted siRNA groups was 21.5% and 26.28%,both higher than normal Lovo's. The cell proliferation of two survivin-targeted siRNA groups was both lower than normal Lovo's. Conclusion The 2 siRNA targeting survivin gene can suppress the expression of survivin mRNA, inhibit cell growth,and make the apoptosis higher. It provides a powerful evidence of carcinoma of colon gene therapy
