Abstract:[Objective]To understand the distribution of genotypes and mutation of hepatitis B virus(HBV) in hepatitis B patients.[Methods]Eighty-eight hepatitis B patients in Shanghai were randomly sampled in 20...[Objective]To understand the distribution of genotypes and mutation of hepatitis B virus(HBV) in hepatitis B patients.[Methods]Eighty-eight hepatitis B patients in Shanghai were randomly sampled in 2010.Nested polymerase chain reaction(PCR) was carried out to detect genotypes of HBV in blood samples of the selected patients.The pre-S/S and C gene regions of HBV DNA were amplified and sequenced.[Results]Among the 88 subjects,52 patients(59.09%) were infected by HBV genotype C and 36 patients(40.91%) by HBV genotype B.The mean age of HBV genotype B group[(38.56±12.55) years old]was lower than that of HBV genotype C group[(44.54±15.72) years old]with significantly statistical difference(P0.05).The positive rate of hepatitis B e antigen(HBeAg) in the HBV genotype B group(66.67%) was significantly higher than that of the HBV genotype C group(28.85%)(P0.05).The mutation rates of C1762 and C1764 situs in HBV genotype C group were 50% and 50.77% respectively,statistically higher than those of the genotype B group which were 27.78% and 25% respectively(P0.05).[Conclusion]The hepatitis B patients in this study are dominantly infected with genotype C.The mutation rates of C1762 and C1764 situs in patients with genotype C were higher than those with genotype B.Strengthened surveillance on genotype and mutation of HBV is required.Read More
