Title: Therapeutic Effect of rmhTNF on Nude Mice Transplanted Tumor Model of Human Gastric Cancer Cell Line
Abstract: Background: Recombinant mutant human tumor necrosis factor (rmhTNF),a mutant of prototype TNF-α,has been reported to have growth inhibition and apoptosis induction effects on human gastric cancer cell line in vitro in our previous study.Aims: To investigate the therapeutic effect of rmhTNF on nude mice transplanted tumor model of human gastric cancer cell line.Methods: Male BALB/c nude mice were inoculated with human gastric cancer cell line BGC-823 subcutaneously to establish transplanted tumor model.The nude mice bearing transplanted tumor were randomly allocated into four groups receiving rmhTNF,TNF-α,5-fluorouracil (positive control) and normal saline (negative control) interventions,respectively.The general status,growth of implanted tumor and its histopathological changes were observed. Results: The success rate of establishing transplanted tumor was 100%.Tumor growth was significantly decelerated in the three interventional groups as compared with the negative control group,especially the rmhTNF group,in which the growth curve of transplanted tumor was depressed markedly,and the tumor inhibition rate was significantly higher than that in TNF-α group and positive control group (83.1% vs.59.8% and 50.1%,P0.01),and the general status was not significantly changed when compared with that before inoculation.In addition,more apoptotic and necrotic cells were seen in tumor tissues of rmhTNF group.Conclusions: BGC-823 cells showed well tumor formation when inoculated subcutaneously in BALB/c nude mice.rmhTNF can induce apoptosis and necrosis in vivo,and has therapeutic effect on nude mice transplanted tumor model of human gastric cancer cell line.
