Title: The change of aquaporin-2 mRNA in the graft of acute rejection rat renal transplantation model
Abstract: Objective To study the change of aquaprorin-2(AQP2) mRNA in the graft of the rat renal transplantation model,especially after the acute rejection.Methods Normal Wistar rats' renals were selected as control group,Wistar rats acting as both donors and receptors were selected as syngeneic control group,and SD rats acting as donors but Wistar rats acting as receptors were selected as allogeneic acute rejection group 〔given cyclosporine A(CsA)〕 and allogeneic immunosuppression group(without given CsA).All rats were male.RT-PCR method was applied to test the change of AQP2 mRNA expression of each group.Results The AQP2 mRNA expression in allogeneic acute rejection group was decreased significantly compared with that of other three groups(P0.05).Compared with control group,the AQP2 mRNA expression in both syngeneic control group and allogeneic immunosuppression group had no significant change(P0.05).Conclusions Downregulation of AQP2 mRNA may reduce reabsorption of water and energy consumption in collecting duct of renal transplant in acute rejection phase,which is beneficial to the recovery of renal transplant function.
