Title: Initiation and Regeneration of Callus in Maize Inbred Lines
Abstract: Using immature embryos of eight maize inbred-lines widely used in Heilongjiang agriculture production as explants to induce embryogenic calluses, the effect of genotype and the concentration of 2,4-D on the induction of calluses was studied. The results shows that culture of maize tissue relies more on genotype; concentration of 2,4-D should be high during initiation, but in the process of subculture, it must be adjusted to callus conditions. Factors that affect differentiation, regeneration and transplanting were analyzed in order to build an active regeneration system for gene transformation.
