Title: Cytotoxicity of Trace Metal Copper in Cisplatin-sensitive and-resistant on Oral Carcinoma Cells
Abstract: Objective:To investigate the influence of copper on cytotoxicity of tumor cells at the cell lever by a series of tests and search a new method to solve drug resistance of cisplatin(DDP).Methods:With MTT assay,different dose of copper acetate was used to investigate the cytotoxicity of copper and DDP on oral tumor cell of Tca8113,Ace-2 and Ace-3 cell lines.The flow cytometric analysis was used to detect the influence of copper acetate on apoptosis of tumor cells and cell cycle.Results:Copper acetate in the concentration of 20~400μmol/L significantly suppressed the survival rate(SR) of three tumor cells line.In the presence of 25μmol/L copper acetate combined with various doses of DDP,tumor cell SR were significantly inhibited than that exposed to DDP alone.The flow cytometric analysis detected that 25μmol/L copper acetate increased the apoptosis of cells exposed to 7.5μmol/L DDP,while decreased the apoptosis under 25μmol/L DDP; it also showed that 25μmol/L copper acetate combined with 7.51μmol/L DDP decelerated tumor cell to move to S phase, while accelerated tumor cell to move to S phase if combined with 25μmol/L DDP.Conclusion:Cu~(2+)enhanced anticancer cell response in low density DDP and changed apoptosis and cell cycle of tumor cells exposed to DDP.
