Title: Optimization of the extraction conditions for flavonoid constituents in spanishneedles herb
Abstract: Objective To optimize the extraction conditions for flavonoid constituents of spanishneedles herb,and to establish a method to quantitatively determine the total flavonoids from the extracts of spanishneedles herb.Methods The total flavonoids in spanishneedles extracts was determined by UV-spectrophotometry.The concentration of ethanol,quantity of ethanol,extraction times and time of extraction were investigated by orthogonal test with the quantity of total flavonoids as a target index.Results The total flavonoids in spanishneedles herb were extracted effectively by ethanol.Linear range of the rutin concentration was 0.004 7-0.023 5 mg/ml,and regression equation was A=28.723c+0.043 4,r=0.999 6(n=5).RSD was less than 5%.The mean recovery was 98.58%.The optimized extraction conditions were 12 times as much as the volume of 80% ethanol for three csucessive extraction and 1.0 h per time.Conclusion The method could qualitatively and quantitatively determine the total flavonoid in spanishneedles herb.The optimized extraction method is stable and efficient.
