Title: Relationship of expression of Fas-L,Caspase-3 and NF-κB and hepatocyte apoptosis in septic rat
Abstract: Objective To investigate the role of Fas-L,Caspase- 3 and NF-κB in signal transduction in heptocyte apoptosis.Methods Thirty Wistar rats were randomly divided into the normal control amd the exp erimental group.Twenty-four of them were subjected to septic model induced by c ecal ligation and puncture (CLP),and they were respectively sacrificed at 2 h,3 h,4 h,5 h after CLP.The levels of expression of Fas-L,Caspase-3 and NF-κB protein in liver tissues and hepatocyte apoptosis were measured at various intervals in liver tissues. Alanine aminotransferase(ALT) and lactate dehydrogenase(LDH) wer e also detected at the same time.Results Expression of Fas-L, Caspase-3 and NF-κB protein in liver tissues significantly increased compare d with the control group.Content of Fas-L,Caspase-3 of the 3-h group approach ed the peak.Content of NF-κB of the 4-h group approached the peak.Meanwhile,s erum ALT and LDH were markedly increased compared with the control group(P 0.01),serum ALT showed a progressive increment,and increment of activity of seru m LDH in the 4-h group was prominently,Index of hepatocyte apoptosis showed a progressive increment.Conclusion The expression of Fas-L and Caspase-3 protein increased with activated apoptosis during sepsis induced by CLP;the activity of NF-κB protein was increased as well, and it might suppres se hepatocyte apoptosis.
