Title: Preliminary investigation on the culturable bacteria diversity in Tianjin Coastal Waters
Abstract: Tianjin is located on Bohai bay which is the largest inner ocean in China.Investigation on the bacteria diversity is of benefit to exploitting marine biological resources and improving the ecological environment of Tianjin coast.On the basis of the geographic features of Tianjin coast,5 sites were selected to collect seawater samples for the examination of culturable bacteria and investigation of population diversity.Bacterial populations of seawater were enumerated by the plate count method.The results showed that plate count values ranged from 0.28×107 to 6.08×107 cfu/L.In order to understand the diversity of culturable bacteria,276 strains of becteria were isolated and cultured from different layers in 5 sites.After PCR of 16S rDNA,27 types of RFLP patterns were distinguished with RsaⅠ,MspⅠ.On the basis of the sequencing of 16S rDNA genes and comparing with the published sequences in GenBank database,27 clonies fell into three groups: Proteobacteria,Irmicutes,Acteroidetes.Most of sequences were 96% identical to the published sequences.The Phylogenetic analysis has been performed by use of 16S rDNA sequencing and RFLP(restriction fragment length polymorphism).
