Methods: observe the direct effects of As on myocardiac contractility by a pattern of Langendorff isolated Wistar rat heart perfusion through determination of hemodynamics parameters including aorta flow (AF), coronary flow (CF), heart rate, arterial blood pressure (AP), left ventricular systolic pressure (LVSP), left ventricular end diastolic pressure (LVEDP), and ±dp/dt max . Results: 10, 20 and 40 mg·L -1 As all increased AP, LVSP,±dp/dt max obviously, showed that As could strengthen the cardiac contractility in a dose dependent manner. The effects immediately weakened after drug was washed out. It was found that the positive inotropic action was inhibited by 0.02 mg·L -1 verapamil. Conclusion: The results suggeste that As may cause positive inotropic effect in isolated working heart in a dose dependent manner, and the inotropic action is calcium dependent.Read More
