Title: Tissue Culture and Rapid Propagation of Ludisia discolor
Abstract: This paper deals with tissue culture of Ludisia discolor by tender stem explants. The results showed the suitable medium of initial was MS medium supplemented with 5.0 mg·L-1 6-BA. The best proliferation occurred on MS medium supplemented with 5.0 mg·L-1 6-BA and 0.5 mg·L-1 NAA and 0.3 mg·L-1 TDZ, the propagation coefficient of adventitious shoots reached 4.92. The best hardening medium was MS medium supplemented with 0.3 mg·L-1 NAA and 1.0 mg·L-1 GA3 and 15% coconut milk, with the average plantlet height of 4.33 cm and the average stem diameter of 0.61 cm. The best rooting for plantlets occurred on 1/2MS medium supplemented with 1.0 mg·L-1 IBA and 15% banana and 0.5 g·L-1 activated carbons, with rooting rate up to 93.0%. The plantlets with acclimatization were transferred to the the mixed matrix [peat soil:vermiculite:pine bark=3:1:1(V/V/V)] and the outplanting survival rate was 87.0%.
