Title: Study on Osteoponitin Antibody Combined with Chemotherapy Drugs in Osteosarcoma
Abstract: Objective:To investigate the effectiveness of osteopontin antibody in an osteosarcoma cell line MG-63,and to investigate the relationship of osteopontin antibody and chemotherapeutic drugs.Methods:An osteosarcoma cell line MG-63 was treated by osteopontin antibody combined with concentration gradients of MTX,ADM,DDP and IFO4,respectively.MTT assay was utilized to detect the cell viability at 0,24,48,and 72 hours after treated.Results:The inhibitions of cell proliferation by chemotherapeutic drugs on MG-63 cell were elevated by osteopontin antibody,which were dose dependent and time dependent.In certain concentration of chemotherapy drugs,there was significant elevation of the anti-proliferation effects by osteopontin antibody treatment.This indicated that osteopontin antibody prompted the chemotherapeutic sensitivity.Our data showed the synergetic effect of osteopontin antibody and chemotherapeutic drugs in the treatment of osteosarcoma.Conclusion:The osteopontin antibody improved chemotherapeutic sensitivity in a synergetic manner with chemotherapeutic drugs in MG-63 cell.The osteopontin antibody is potentially applicable in clinical practice for osteosarcoma.The osteopontin is a prospective target in medical treatment of osteosarcoma.
