Abstract:OBJECTIVE: To observe the effect of poly(ADP-ribose) polymerase(PARP) inhibitors on the growth inhibition by cisplatin in the Cis-sensitive and Cis-resistant lung cancer cell lines.METHODS: The effect...OBJECTIVE: To observe the effect of poly(ADP-ribose) polymerase(PARP) inhibitors on the growth inhibition by cisplatin in the Cis-sensitive and Cis-resistant lung cancer cell lines.METHODS: The effect of proliferation inhibition of the combination of PARP inhibitors with cispaltin and that of the single drugs were detected by MTT assay.The difference of expression of PARP-1 and PARP-2 between the Cis-sensitive and Cis-resistant cell lines,together with the formation of Poly(ADP-ribose)(PAR) following the exposure of cisplatin and the inhibition of PAR by PARP inhibitors were analyzed by western blot analysis.RESULTS: The expression of PARP-1 between the parent cisplatin sensitive cell lines PC9,PC14,SBC3 and their sub cisplatin resistant cell lines was significantly different(P=0.028,0.013,0.008).After exposure of 60 minutes to cisplatin,PARP was activated rapidly and the formation of PAR was the highest at 40 minutes and could be inhibited by DPQ in the lung cancer cell lines(P0.05,t0).PARP inhibitors did not potentiate the effect of growth inhibition of cisplatin in all lung cancer cell lines.CONCLUSIONS: PARP inhibitors brings effects of no potentiation of growth inhibition of cisplatin in all the Cis-sensitive and Cis-resistant lung cancer cell lines,which suggests that the different expression of PARP-1 is not related with cell lines whether sensitive to cisplatin or not.Further investigations are still needed to observe the effect of PARP inhibitors on proliferation inhibition by other DNA damage chemotherapy agents in lung cancer cell lines.Read More
