Title: THE MEASUREMENT OF 17β-ESTRADIOL RECEPTOR IN HUMAN ENDOMETRIUM
Abstract: On purpose to study the female reproductive physiology, a modified method for the measurment of oytoplasmic and nuclear 17β-estradiol receptors in human endome-trium was established and performed on 43 normal women of 24-25 years old. According to the method of Francis Bayard, the amount of oytoplasmic protein was measured by Hartree. The method of Burton was used to identify the DNA content in the nuclei.The amount of the receptor of cytoplasm was expressed by fmol/mg protein and the receptor of nuclei was expressed by fmol/mg DNA.Scatchard plot analysis was made, the Kd (equilibrium dissociation constant) of 17β-estradiol is about-10-10M. Best result was obtained with the optimum temperature of 25℃.We succeeded in identifying specific estradiol receptor in normal endometrium of 43 women. The amount of estradiol receptor concentration was highest during the proliferative phase and was very significantly lower in the second half of the menstrual cycle. The result is similar to that reported by 0 Levy, et al in 1980.
