Abstract:Objective To verify the interaction between coiled-coil structure-containing PML domain(PML-C) and CNPY2 protein by in vitro and in vivo tests.Methods Yeast AH109 cells were co-transformed with plasmi...Objective To verify the interaction between coiled-coil structure-containing PML domain(PML-C) and CNPY2 protein by in vitro and in vivo tests.Methods Yeast AH109 cells were co-transformed with plasmids pACT2-CNPY2 and pGBKT7-PML-C,based on which the interaction between CNPY2 and PML-C was verified by yeast two hybrid technique.HEK293 cells were co-transfected with the constructed recombinant plasmids pCMV-HA-PML-C and pCMV-Myc-CNPY2 and determined for expression of CNPY2 protein by Western blot.The interaction between the two proteins was verified by co-immunoprecipitation in vitro.Results Blue positive clones were observed in yeast AH109 cells transfected with plasmids pACT2-CNPY2 and pGBKT7-PML-C.The lysate of HEK293 cells co-transfected with plasmid pCMV-HA-PML-C and pCMV-Myc-CNPY2 showed specific binding to mouse anti-c-MYC monoclonal antibody.The protein complex which showed interaction with PML-C was detected by co-immunoprecipitation.Conclusion The interaction between PML-C and CNPY2 protein was proved by yeast two hybrid assay and co-immunoprecipitation.Read More
